Single-Cell Epitope mapping using Antigen-libraries and single-cell secretion assays
This project uses droplet based microfluidics to encapsulate single-cells for antibody capture, characterization and B cell isolation. The current project involves cloning and expression of novel antigen into an antigen-library for antigen mapping at single-droplet resolution. The project is suited for Master projects and master thesis (>3 months) and involves, cloning, validation, expression analysis as well as single-droplet verification. Finally, antibody secretion experiment are planned to isolate single-antigen binders and B cell sequencing.
Current antigens revolve around the SARS Cov2 Nucleocapsid protein, RBD and envelope proteins to assess antibody cross-reactivity.
