A microfluidic platform for osteogenesis in a 3D macroporous culture
Structural and functional models of human tissue, which enable research at high throughput within small volumes, are of great interest. A promising approach consists of on-chip cultures. Microfluidic chips can replace or complement classical 2D cell culture by a more dynamic 3D approach. For example, bone is a highly dynamic, strongly vascularized 3D tissue system. Therefore, a 3D approach is optimal for an accurate in vitro model. As a target organ for many drugs, a more precise replica of this specific tissue would facilitate the investigation of a plethora of therapeutics. Therefore, this platform tries to tackle some of the obstacles that are currently faced. This platform simulates a stiff environment, without preventing proliferation. The material is fully synthetic and therefore controllable and more reproduceable. The system is further capable of lateral perfusion. The overall goal is a highly tuneable system, which can imitate various environmental conditions, with low labour work and a simple readout.
What we offer:
As this project is carried out as a collaborative project between the Laboratory for Cell and Tissue Engineering at the University Hospital of Zurich and the Macromolecular Engineering Lab at ETH Zurich, you will become a member of two labs and therefore get insights into many different research topics and meet a lot of new people.
You will learn:
Cell culture of human mesenchymal stem/stromal cells and endothelial cells
Production and handling of microfluidic chips
Production and handling of microgels
Immunostaining, imaging and image processing
RT-qPCR and MORE! :)
What we are looking for:
Students looking for an exciting and interdisciplinary master thesis project (preferred) or master internship (no shorter than 3 months) are welcome to apply. Experience in one of the areas mentioned above would be an advantage but not strictly required, especially in the case of a master thesis project, where we have more time to learn it all together! :)
Ideally, you would be able to start as soon as possible so we have an overlap with the current student! Later starting times could be arranged, however, upon discussion with us :)
