Responsive nanogels for therapeutic delivery
There are a wide variety of peptides, cytokines, and proteins which offer offer therapeutics advantages but have a limited use because of the detrimental side-effects. One of the possible strategies for using these drugs in a clinical setting is to develop biocompatible materials which localize the therapeutic treatement to the target site allowing less side effects, therapeutic protection, and increased doses. In this project we develop a functional nanogel platform which allows therapeutic protection and local delivery. These nanogels are designed for keeping the therapeutic linked to the material until they are embedded in a reductive environment, such as the tumor microenvironment. The presence of a reducing agent such as glutathione would cleave the therapeutic linker thus releasing the therapeutic at the target site.
Keywords: Chemical polymer functionalization, linker design, nanoparticles, hydrogels, drug delivery, cancer, antimicrobial drugs
We are looking for a Chemistry / Chemical Engineering student for Bachelor or Master Thesis. The project would involve chemical syntheses to make the hydrogel precursors and the glutathione-cleavable linker. Subsequently, if drug release experiments show that our nanogels work, then we will test them on living cells with simple biological experiments. We will learn many of the techniques described here together.
