Open OpportunitiesIn our lab we are analysing droplet-microfluidic generated multi-spectral images of single-cell secreting multiple protein simultaneously. As multifunctionalities of immune cells are understudied we are aiming at developing further data science tools for image analysis, cell detection, multi-functionality read-out and data visualization. This student project aims at building on previous data science and software development projects which have developed a comprehensive pipeline to analyse large fluorescent imaging files from single-cell secretion experiments inside droplets. The software includes droplet detection and tracking, cell identification and scoring, protein secretion read-out and classification, grouping and subpopulation data analysis together with plotting of visualisation tools. pre-loaded PCR test kits that allow for simpler and faster sample preparation steps. In 2020 our
devices have been used by more than 250 laypeople for the rapid detection of COVID-19 in a field
study that we run at ETH Zürich, and our systems are currently being tested at the Swiss Tropical and

| This project uses droplet based microfluidics to encapsulate single-cells for antibody capture, characterization and B cell isolation. The current project involves cloning and expression of novel antigen into an antigen-library for antigen mapping at single-droplet resolution. The project is suited for Master projects and master thesis (>3 months) and involves, cloning, validation, expression analysis as well as single-droplet verification. Finally, antibody secretion experiment are planned to isolate single-antigen binders and B cell sequencing.
